Sequential Observation of Experimental silicosis


Abstract
In order to observe the morphological changes of the lung in the development of experimental silicosis sequential, seven groups of five rats weighing 200gm were injected intratracheally with 0.5§¢ equeous suspension of 50§· of free silica dust. Each group of animals from group 1 to 7 were sacrified 1,3,5,10,25,50, and 100 days after intratracheal injection of dust respectively. The lung tissues of the rats were prepared into slides, fixed and H-E stained.
The microscopical findings at the various stages of the experimental silicosis were as follows:
1. The early changes in the lung were the infiltration of monocytes on the alveolar and bronchiole wall, and emphysematous changes. The more severe changes were observed as the observation period extended.
2. The granulomas developed scarcely in the 5th days group on alveolar and bronchiole wall.
3. The mild fibrosis in the alveoli started from the 10th day, and the severity of fibrosis gradually increased, as the observation period extended.
4. The typical silicotic nodules appeared only the latest stage (the 100 days) of this experiment.
